2 Figure 1 p53 inactivation at initiation step in liver carcinogenesis. Mice with liver-specific p53 disruption developed HCC harboring chromosomal instability and bilineal features. Both mature hepatocytes and liver stem cells purified from p53−/− livers are able to give rise to HCC tumors with stem cell-like markers. Additional inactivation of the retinoblastoma (Rb) pathway participates in tumor development and increases its agressivity.
