Combined inhibition of Wee1 and PARP causes persistent radiation-induced DNA damage. AsPC-1 (A, B) and MiaPaCa-2 (C, D) cells were treated with AZD1775/olaparib for 1 hour pre- and for the indicated times post-radiation (RT) or mock-radiation. At each time point post-radiation, cells were fixed for flow cytometric analysis and analyzed for γH2AX positivity and DNA content. Data shown are the mean percentage of γH2AX-positive cells ± SE from n = 3 to 4 independent experiments.
