Activation of Vγ9 T cells by three distinct stimuli is CD277 dependent. A total of 1 × 105 PBMCs (A and B) or 1.5 × 105 leukocytes (C and D) were stimulated with 2 μM of ZOL, 100 ng/ml agonistic anti-CD277 mAb 20.1, or 1 nM of HMBPP in the absence (A and C) or presence (B and D) of 50 ng/ml IL-18 and absence or presence of 5 μg/ml antagonistic anti-CD277 mAb as indicated. Control IgG1 used in other experiments did not have any effect. All cultures were supplemented with 50 U/ml IL-2. The absolute number of viable Vγ9 T cells per microculture was measured by SCDA after 7 d. Expansion ratios were calculated by dividing the number of Vγ9 T cells on day 7 by the calculated number of Vγ9 T cells present at the initiation of cell culture.
