Dcr-2 is required for Toll signaling in Drosophila S2 cells. (A and B) Cultured S2 cells were transfected with dsRNAs against the indicated genes. Total RNA extracts were prepared from the cells and detected via Northern blots using the indicated probes. (C) Cultured S2 cells were transfected with dsRNAs against the indicated genes and treated with Lys-PGN (100 μg/ml; lanes 2 to 5) or water (lane 1) for 20 hours. Total RNA extracts were prepared from the cells and detected via Northern blots using the indicated probes. (D and E) Cultured S2 cells were transfected with dsRNAs and treated with Lys-PGN as indicated. Total RNA extracts were prepared for qRT-PCR assay of Dros (D) or Dipt (E) mRNA (normalized to Rp49; n = 3; *P < 0.05, two-tailed Student’s t test; error bars, SD).
