Effects of rapamycin, cycloheximide, and an MAP kinase inhibitor (U0126) on BDNF-mediated increase in Arc and CaMKII in acute hippocampal slices. Effects of rapamycin, cycloheximide, and an MAP kinase inhibitor (U0126) on BDNF-mediated increase in Arc and CaMKII in acute hippocampal slices. A, B, Acute hippocampal slices were pretreated with DMSO (0.01%), rapamycin (1 μm), cycloheximide (25 μm), or U0126 (5 μm) for 30 min and then incubated with BDNF (100 ng/ml) for 30 min. Slices were homogenized and processed for Western blot for Arc (A) and CaMKII (B). Data are normalized to the levels of actin and are expressed as ratio over control values; they are means ± SEM from three to five independent experiments. *p < 0.05 (compared to vehicle); #p < 0.05 (compared to BDNF alone; two-way ANOVA).
