A, TSG-6 staining in human prostate. Human prostate tissue array containing different grade tumors with paired normal prostate was stained with TSG-6. TSG-6 was used at a dilution of 1:100 in PBS and incubated overnight at 4°C. Immune complexes were revealed using a universal secondary antibody (100 μL for 30 minutes) followed by chromogen as described in Materials and Methods. Left, little or no TSG-6 staining in the cancerous tissue; right, TSG-6 expression in normal prostate (P = ). Statistical significance of the data was determined using ANOVA and bootstrap methods as described in Materials and Methods. B, variability of TSG-6 expression in human prostate tumors. Tissue array containing human prostate tumors from different Gleason grade were incubated with a protein blocking serum-free reagent for 60 minutes to block nonspecific binding. TSG-6 was used at a dilution of 1:100 in PBS and incubated overnight at 4°C. Immune complexes were revealed using a universal secondary antibody (100 μL for 30 minutes) followed by chromogen.
