Effect of bevacizumab on the proliferation of A2780 cells. A, Western blot analysis of VEGF expression. A2780 cells were incubated for 16 h under nonhypoxic (20% O2) or hypoxic (1% O2) conditions in the presence of 5% fetal bovine serum. Cells were harvested, and equal amounts (30 μg) of protein were subjected to SDS-PAGE, blotted, and incubated with either anti-VEGF or anti–β-actin antibody. B, A2780 cells were treated with the indicated concentrations of VEGF with or without bevacizumab in the presence of 5% fetal bovine serum. Cell viability was assessed by MTS assay. Columns, mean; bars, SD. A statistically significant difference is indicated by asterisk (**, P < 0.05). Experiments were repeated in triplicate with similar results, and representative results are shown.
