PARP1 suppresses the transcription of PD-L1 in cancer cells. A, Heat map showing the mRNA fold change when PARP1 was silenced. The transcription of immunosuppressive factors was analyzed by qRT-PCR in the human ovarian cancer cell line OVCAR8 when PARP1 was downregulated by siRNA (siPARP1). B, The PD-L1 mRNA expression was measured in OVCAR8 and SKOV3 cells transfected with siRNA-NC and siPARP1. C, PD-L1 protein expression with PARP1 silenced. The PD-L1 protein expression was determined by Western blot in OVCAR8 and SKOV3 cells when treated with PARP1 siRNA-NC and siPARP1. D, Cell-surface PD-L1 expression with PARP1 silenced in both OVCAR8 and SKOV3 cell lines. Cell-surface PD-L1 was measured by flow cytometry in OVCAR8 and SKOV3 cells when the cells were treated with siRNA-NC and siPARP1. Cells were assessed for PD-L1 or mouse IgG control antibody (IgG).
