Cells expressing mutant NADK-I90F exhibit low ROS and elevated NADP and NADPH levels. Total NADP and NADPH (NADPt) and NADPH levels were determined in normal pancreatic ductal cells (HPDE); pancreatic ductal adenocarcinoma cells (BxPC-3 and Panc-1); or cell lines expressing either the vector control, NADK, or NADK-I90F using the NADP/NADPH-Glo assay (Promega; A). ROS levels were assayed in normal pancreatic ductal cells (HPDE) or pancreatic cancer cells (BxPC-3 and Panc-1; B). Expression levels of NADK and NADK-I90F were determined (C).
