Preparation of pooled plasma as a control for coagulation tests

Best for large hospitals which have a busy blood bank (with a facility of -80 o C freezer) Preparation of pooled plasma

Healthy donor’s plasma segments are collected and immediately frozen at -80º C. All these segments are thawed at 37º C, cut and pooled in a plastic container. This pooled plasma is centrifuged at 4000 rpm for 10 minutes to obtain platelet poor plasma. Platelet count is analyzed for representative pool. Platelet count should be <10000 ×10 9 L. This pooled plasma is aliquoted in vials and immediately stored at -80º C. Pooled plasma which has been prepared is analysed on ACL analyzer for PT, PTT & PT derived Fibrinogen. This pooled plasma is used as a Normal control with every batch of samples. Pooled plasma – Normal control

Pooled plasma -Abnormal control Pooled plasma diluted with phosphate buffer saline ( pH-7.2 ) (1:3) This may be used as an abnormal control.

PT, PTT & derived Fibrinogen readings of control are monitored for every batch. The mean of 21 readings are taken and it is programmed in reference data of ACL analyzer for PT, PTT & Fibrinogen.
