Learning Goals G 1 Understand that signaling pathways are ordered and specific to maintain normal cell function. G 2 Understand that cells regulate their function and behavior by a complex set of molecular interactions. G 3 Understanding the importance of signal modulation in affecting cell behavior. 2

Learning Objectives O 1 - Illustrate and identify individual components of a cellular pathway. O 2 - Be able to assign locations for given components/roles of a signaling pathway. O 3 - Predict outcomes of inhibitors/ activators that target components of a pathway and predict the effect of various interventions in a signaling pathway. 3

Build a Pathway, Part Two Imagine that our virtual cell is generating excess heat in the absence of cold stimulus! a)Propose one plausible hypothesis to account for this observation. b)How would you test your hypothesis? Please focus on what you would test and why, but not the method. c)Design a treatment targeted at one or more components of the pathway that would normalize heat production by the cell. d)Be prepared to present your results to the class for discussion! (formative assessment) 6

Recap: Learning Objectives Illustrate and identify individual components of a cellular pathway. Be able to assign locations for given components/roles of a signaling pathway. Predict outcomes of inhibitors/ activators that target components of a pathway and predict the effect of various interventions in a signaling pathway. 8

Summative Assessment Our virtual cell is not generating heat in presence of a cold stimulus. The level of receptor-generated second messenger is elevated, but transcription factor is not phosphorylated. – Draw a generalized model of the signaling pathway. – What hypothesis would be consistent with both of these observations? – How would you test your hypothesis? What would you test and why? – Design a treatment that would normalize heat production by the cell. 9
