2. Step 1: Extract DNA from Banana
Follow the instructions listed on your handout.
(Steps # 1-9)
Materials on the cart: Amt you need
-Shampoo teaspoon
-Salt pinches
-Vial w/chilled 91% alcohol 1 vial/pair
-De-ionized water ml
-Banana mixture 3 teaspoons
**Important notes & Reminders:
-Note the appearance & describe what you see.
-Rinse out your vials after you have made your observations
-Answer questions # 1-4

3. Follow up Lab: Extracting Your DNA http://www. pbs
Supplies are in the back of the room.
1) Pick up a dixie cup w/ salt water (2 tsp)
**TWO teaspoons salt in one quart (liter) Dist. water
2) Swish around in your mouth thoroughly for 30 sec.
3) Spit back into the cup
4) Pour the contents of the cup into the vial with soap solution (Woolite or Palmolive 3:1 ratio - water & soap)
5) Gently rock the vial back & forth (3 min)
6) Take one pipette full of chilled alcohol (95% Ethanol) and slowly add it to the vial while tilted making sure that they don’t mix.
7) Let stand for 1 minute
8) Gently mix the alcohol into the soap layer with a glass rod.
9) DNA should precipitate (whitish) –Be patient
