Uncouplers prevent a H+ gradient from forming by allowing H+ to diffuse through the membrane (instead of ATP synthase). This prevents ATP synthase from harnessing the energy of the ETC and thus, it cannot phosphorylate ADP to make ATP. Cells continue to consume O2 and food to be broken down in Glycolysis, the TCA, and the ETC. Therefore, the other energy captured from glucose is lost as heat (instead of being used by ATP synthase). -Loss of weight -Excess sweating -Increase in body temperature -Protein denaturation = no metabolism = death
