Sterile Technique

Important to keep work area as clean as possible Prevents introduction of other microorganisms from the environment into your culture The techniques used are sterile techniques

Sterile Technique Wash with a surface disinfectant. The most commonly used are: 1.10% bleach (recommended by the CDC) 2.85% ethanol Aseptic technique – killing microbes with heat

Sterile Technique No strong air current Use a UV bulb in a fluorescent light fixture to surface sterilize area clean and sterilized all glassware, pipettes, spatulas, and test tube (culture) racks You can purchase sterile equipment

Sterile Technique Wash hands after you finish cleaning and put on a pair of sterile disposable gloves before you begin. Once everything is clean and sterile, don’t contaminate the work area by placing “dirty items” such as pencils, pens, notes, or books in the sterile work area

Media Sterilization There are two reliable methods used to sterilize microbial culture media: 1.autoclave 2.pressure cooker

Autoclave Heat and pressure Run for 3-18 minutes (15 for us) heat is usually set between 121 and 134 degrees Celsius The intense heat kills bacteria, viruses and other organisms. Heat needs to circulate around objects Precautions An autoclave will not work to sterilize some items, such as certain plastics, because they can be melted or damaged
