3 BIOC 455: Course Content Dipeptide analysis (2 lab periods) Determination of the amino acid sequence of a dipeptide Photometry (1 lab period) Colorimetric quantitation of total protein concentration. Study of the properties of β-galactosidase (4 lab periods) Kinetic assays of β-galactosidase Study of carbohydrates (1 lab period) Enzymatic and chemical determination of carbohydrate unknowns Purification of Glutamate Oxaloacetate Transaminase from pig heart muscle (5 lab periods) Three step purification of an active enzyme from a natural source.

4 BIOC 455: Course Content Study of the interaction of RNA polymerase subunits. (1 lab period) Affinity pull-down of separately expressed RNA polymerase subunits from Methanococcus jannaschii. Study of the properties of aspartate transcarbamylase (1 lab period) Kinetic assays of the allosteric enzyme ATCase Use of the ELISA in immunology (3 lab periods) Determination of the titers of purified sera, unpurified sera, and monoclonal ascites. Characterization of Erythrocyte Membranes (2 lab periods) Extraction of protein and lipid fractions of pig red blood cells. In vitro transcription/translation (2 lab periods) Coupled system to produce isotopically labeled protein product
