Activity: Make Your Own Primers Virginia iGEM Workshop #3 High School Education Series DON’T FORGET! 18-22 base pairs 50-60% GC Content Wallace Rule: 4 degrees per C or G, 2 degrees per A or T Melting temperature of the primer. Subtract two degrees for annealing temperature.
