1 Lox Site Random Non-coding DNA (1-2kb) Stop Codons Antibiotic Resist Cre Pops Blocker This device will prevent Pops from a promoter reaching target genes. Activation of the Cre Recombinase genes will remove the stop codons and allow Pops to pass through this part. Removal of the stop codons also allows transcription of an antibiotic so the activated bacteria can be selected. The restriction sites allow the part to be ligated into a system in the normal fashion. This part can only be activated once and will remain activated forever. It should be 100% efficiant at stopping Pops passing through it until activated. No other molecules are required for any of the steps, simple. A separate induceable Cre plasmid must be inserted to produce the cre recombinase Antibiotic Resist

2 How Cre Works Cre is under the control of a promoter, there are commercially available Cre containing plasmids
