General Directions for all Tests
Before beginning, make predictions for all tests. Label the test tubes 1-7 Note do not cross contaminate tubes between tests. Use the same substance in the same tube for each test 3. Place a small amount of each substance in the tube (approx. 1 cm) 4. Add 8-10 drops of each indicator to the tube and record your results.

Lipid Test – Sudan III Stain
This indicator will turn bright red or pink in the presence of lipids.

Protein Test Biuret reagent will turn from blue to violet in the presence of proteins. *Caution* this indicator is corrosive and will burn your skin!

Iodine Complex Carbohydrate Test
Complex carbohydrates or polysaccharides are tested for using Iodine. It will turn from a light brown to a dark blue or black.

Simple Sugar Test Monosaccharides and disaccharides are indicated when the sample treated with Benedicts solution is heated. If simple sugars are present it will turn from blue to green, yellow, orange, or red.

pH test. Apply a small amount of each substance to pH paper and compare to the provided color chart to determine pH.
