2 Screening of wheat cultivars for BSMV infection.
Bezosztaja M M M Mv Táltos Chul Mv Hombár HvParis Axminster Online Resource Fig .1 Screening of wheat cultivars for BSMV infection. 5 different wheat cultivars was infected with BSMV and the infection efficiency was compared using a barley cultivar as a reference. Northern blot analysis of RNAs extracted at 15dpi from systemic 3rd leaves of the infected plants were hybridized with radiolabelled DNA probe generated from BSMV ɣDNA.

3 AG SH SC Online Resource Fig. 2 Typical stages of powdery mildew development used for sorting in infection test. Spores forming appressorial germ tubes (AG), spores showing early phase of elongating secondary hyphae (SH) and spores forming sporulating colonies (SC) at 4 dpi. The size bar is 0,1mm .

4 Online Resource Fig.3 Result of powdery mildew infection after effective VIGS. Coloumn diagram representation of the infection with BSMV:MLOA and BSMV:0 as control. Leaf segments of the infected plants at 15dpi were infected with powdery mildew (mixed strain). The infected leaves were stained with Trypan blue after 4 days, and the spores on the leaf were classified according to their germination stages: non germinating (NG) spores, spores forming appressorial germ tube (AG), spores showing early phase of elongating secondary hyphae (SH) and spores forming sporulating colony (SC). The upper panel shows the percentage distribution of the different stages as a column diagram (Samples 1 and 22 on Fig 3.), while the panel below is a picture of a typical powdery mildew infected leaf after Trypan Blue staining . The size bar is 0,25mm.
