2 Adipocytes play important roles in synthesis, storage, and hydrolysis of nutritional energy. They store triglycerides during periods of caloric excess, but mobilize this reserve during caloric insufficiency, thereby they maintain whole-body energy homeostasis in animals. Adipogenesis is the process of cell differentiation of pre-adipocytes (derived from mesenchymal stem cells (MSCs)) into adipocytes. Throughout development, adipocytes acquire a full complement of enzymes and accessory proteins to carry out de novo lipogenesis, lipoprotein hydrolysis, intracellular fatty acid transport, and lipolysis. Adipogenesis is tightly regulated by hormonal and growth factor signaling, stimulating pre-adipocyte differentiation and comprises regulation of genes involved in all aspects of molecular events such as cell morphology, storage and release of triglycerides, glucose metabolism, and energy homeostasis. Extensive research on adipogenesis has implicated dysfunctioning adipocytes in pathological states such as diabetes, atherosclerosis, cancer and cardiovascular disease. Further research can identify regulators of adipogenesis and potentially find ways to prevent and treat diabetes and other obesity related diseases. 3 Service Details: Total triglyceride differentiation assays are performed in a 96-well or 384-well format using subcutaneous preadipocytes. Compounds are tested for their ability to increase triglyceride accumulation during the differentiation process compared to the positive control of complete differentiation medium. Triglyceride is quantified using a coupled enzyme colorimetric reaction and comparison to a standard curve. DMSO vehicle is a negative control and PPARα agonists are the positive control. Standard service includes 10 customer-defined compound concentrations each tested in triplicate. Customer should provide test concentrations and optimal sample for the service is a compound dissolved in PBS or culture medium.
