Tashkent Medical Academy Department: Public health and hygiene
2
TOPIC: TSN. Familiarization with the activity of the work of sanitary and bacteriological laboratory CSSES Tashkent. Subject: Communal Hygiene
3
Presentation of workshops on environmental sanitation for the 6th year 11-12 - Semester
4
1 Familiarization with normative legal documents in the field of sanitary and bacteriological laboratory 2 Familiarize themselves with the structure of sanitary and bacteriological laboratory CSSES g Tashkent. 3 Seize the methodic of sanitary and bacteriological studies. 4 Learn how to make health report on the results of laboratory research. 5 Learn to develop preventive measures to protect public health.
5
The student should know: The meaning and legal rules governing the relationship of man to man, society and the environment. Apply the basic provisions of each document in its significance. Features of the organization and conduct of sanitary and epidemiological measures to mitigate the adverse effects of risk factors on health and the environment.
6
The student should be able to: To carry out the current sanitary supervision on controlled utility facilities. Justify their actions in solving the problem. Carry out sanitary and bacteriological studies and appropriate accounting and reporting documentation.
7
Work in the microbiology laboratory of medical institutions is carried out to cause infectious disease pathogens. Therefore, to protect against infection personnel must strictly follow the rules.
8
1 All employees are required to work in medical gowns, caps and spare shoes. Log in lab coats without strictly forbidden. Where necessary, working wear face mask of gauze. Working with particularly dangerous microorganisms is regulated by special instructions and carried regime laboratories. 2 The laboratory is forbidden to smoke and eat 3 The workplace must be kept in good order. Personal belongings of the employee should be stored in designated areas. 4 In case of accidental contact with infectious material on the table, floor, and so on. This place should be carefully handle disinfectant solution. 5 Storage, surveillance cultures of micro-organisms and their destruction must be carried out in accordance with special instructions. All cultures of pathogens Register comfort in a special journal. 6 At the end of work hands should be thoroughly cleaned and, if necessary, treat disinfectants.
9
Principles of organization and equipment of bacteriological, virological and serological laboratory
10
Laboratories usually placed in several areas, which, depending on the workload and purpose occupy a certain area. Each lab includes: a) boxes to work with specific groups of bacteria or viruses; b) facilities for serology, culture media preparation, sterilization, washing dishes; c) vivarium with boxes for healthy and experimental animals; g) registry for receiving and issuing analyzes.
11
Laboratories are equipped with the following equipment: biological immersion microscope with additional devices, fluorescence microscope, thermostats, instruments for sterilization (autoclave drying cabinet convolutors)
12
pH meter,Apparatus for the production of distilled water (distiller)centrifuges, technical, analytical balance, apparatus for filtering, water baths
13
refrigerators apparatus for the manufacture of Ventoux - marlevyh plugs set of tools glassware (tubes flasks, Petri dishes, mattresses, vials, ampoules, and Pasteur pipettes), and others.
14
Apparatus for cultivating microorganisms, and other microbiological sterilization purposes. Thermostat. The unit in which the temperature is kept constant. The optimum temperature for reproduction of many microorganisms 370S. thermostats are dry air and water. Used for the cultivation of microorganisms. Mikroanaerostat. Apparatus for cultivating microorganisms under anaerobic conditions. Oven (oven Pasteur). Designed for the sterilization of laboratory glassware and other materials. Autoclave. Designed for steam sterilization under pressure. In the microbiological laboratory autoclaves used different models.
15
The Fridge. Use in microbiology laboratories for storage of cultures of microorganisms culture media, blood, vaccines, sera and other biological active agents at a temperature of about 40C. To maintain a temperature below biologics 00C using low temperature refrigerators, in- which the temperature is maintained below 200C and. Centrifuges. Used for deposition of microorganisms, erythrocytes and other cells for separating heterogeneous liquids. In laboratories using centrifuges operating at different speeds. A device for counting colonies. Semi-automatic meter equipped with a needle with a spring device. Easy push the needle on the bottom of the Petri dish area corresponding to the position of the colony leaves mark on the glass. In this case, the holder rises, the circuit is closed and the counter incremented by one.
16
Dark-field microscopy. Dark field microscopy links based on the phenomenon of light diffraction with a strong side lighting suspended in a liquid of tiny particles. This is achieved by using paraboloidal or kardioid- condenser, which replaces the conventional condenser in a biological microscope.
17
Phase-contrast microscopy. Based on changes in phase transformation occurring during the passage of a light wave through the so-called phase objects, changes in amplitude that caught the eye. With the phase contrast adaptation phase changes of light waves passing through the object are converted amplitude and transparent objects are visible under the microscope. At the same time they acquire a high-contrast image, which can be positive or negative. Positive phase contrast image of the object is called the dark in a bright field of view, the negative phase kontrastom- light image of the object on a dark background.
18
Fluorescent (or fluorescence) microscopy. based on the phenomenon of photoluminescence. Primary Fluorescent staining observed without object, secondary occurs after special staining Fluorescent microscopy as compared with the conventional methods has a number of advantages: possibility of studying living microorganisms and their detection in the material at low concentrations due to the high degree of contrast. In laboratory practice, fluorescence microscopy is widely used for the detection and study of many microorganisms.
19
Sources of waste water and their features methods of mechanical cleaning wastewater. principle of the circular tanks device and small sewerage scheme Biological methods purify household wastewater. Natural and artificial construction of biological wastewater treatment. Hygienic requirements conditions descent wastewater. Methods of treatment of industrial wastewater. Principles of Operation zagnevatelya settlers. Evaluating the effectiveness of treatment facilities for technical and hygienic parameters.
