Instructions This activity was developed as part of the RCSB Collaborative Curriculum Development Program 2014. Work on one sheet at a time to avoid mixing up the strips. The antibody model contains 2 light chains and 2 heavy chains. There are 2 sheets for each light chain and 4 for each heavy chain. For each sheet: 1.Cut out each strip along the dotted lines: Note that the shaded regions labeled la, lb, etc. should be included in the strip Cut out the Disulfide Linkage strips for modeling disulfide bridges 2.Make the immunoglobulin domains: There are 2 immunoglobulin domains in each light chain (A and C) and 4 domains in each heavy chain (B and D) Tape together the cut out strips into a long strand for each domain to represent the polymer backbone The labels la, lb, etc. on the white background should be taped on top of corresponding labels on the shaded background There should be 12 such long strands in all. Assemble the immunoglobulin domain by taping together the strands side by side as you see on the picture. Align the grey area between the strands. Make sure that the beta strand arrows point up and down alternately. Use the red labels and create clusters as follows: 1.Chain A & C: 1A–1D, 1E–1I, 2A–2D, and 2E–2G 2.Chain B & D: 1A–1D, 1E–1I, 2A–2D, 2E–2G, 3A–3D, 3E–3G, 4A – 4D, 4E–4G 3.Assembling the 4 polymer chains Tape together the 2 domains of the light chain at the label lh Tape together the 4 domains of the heavy chain at the labels hi, hr and haa 4.Disulphide linkages Fold down both hatched regions (marked AL1, AL2, etc.) on the strips representing the disulphide linkages. Identify the round labels marked AL1 and AL2 on the Chain A. Place the corresponding disulphide strip on the back of the printed side aligning the hatched regions with the round label. Repeat this for each immunoglobulin domain. Identify the dotted round labels AL and BH on the model and connect them with corresponding disulphide strip. Tape the printed side of the hatched regions on top of the dotted round label. Repeat for the strips marked CL-DH, BHA-DHA, BHB-DHB, BHC-DHC. Suggestion: This model can be made as a group project. Assign each sheet to a different individual to cut and assemble the domain. The 12 different domains in the antibody can be assembled together and all the disulfide bridges put in place as a group. The completed antibody model should look like the picture shown on the next slide
