NCRPIS Imaging Methods 40,000 ft. view of how NC7 processes images –1. Acquire Image –2. Load into Photoshop Insert Meta-Data into image Save full-size TIFF (20 Mb) Save full-size JPEG (0.5 Mb) Save thumbnail JPEG (20Kb) –3. Upload to GRIN Insert Voucher record Insert Observation record Insert Inventory Action record Upload (FTP) JPEG images GRIN Photoshop add-on GRIN Image Loader 1. 2. 3. Full-size JPEG Thumbnail JPEG Full-size TIFF CD/DVD Archive

NCRPIS Imaging Methods Image File Name Protocol Example: PI_123456_00ncai01_SD_ER_LS_2000-01-21_01.jpg ACP (Accession Prefix) ACNO (Accession Number) IVS (Inventory Suffix) IVT (Inventory Type) 2-Character Botanical Part (GRIN) 2-Character Curator Defined Date Image was Acquired Sequence Number of Image Highly functional and concise naming convention. Can be a bit difficult to manually enter – thus the argument for automation.

NCRPIS Imaging Methods Adobe Photoshop JAVA Script Automation File Name: PI_123456_00ncai01_SD_ER_LS_2000-01-21_01.jpg Automated by using the Tyvec field tag containing a barcode of the accession and inventory suffix. Automated by choosing radio buttons and checkboxes in a JAVA script dialog box inside Photoshop Meta-data entry: Automated by choice of radio button and checkboxes in JAVA Script dialog box inside Photoshop. TIFF, Fullsize JPEG, and Thumbnail JPEG: Automated by choosing the Save button in the JAVA Script dialog box inside Photoshop.

Embedded data in the image (from Photoshop): Color palette – storing the color palette with the image allows our color calibration of the scanners to be preserved thus ensuring the original colors of the image are preserved regardless of what machine views the image Text meta-data – The text that we currently store with the image follows: –Document Title = PI_613086_02ncai01_SD_ER_TX_2002-12-02_01 –Author = USDA/ARS/NCRPIS - Iowa State University - Maize Staff –Description = »Maize increase ears. »Captured in TIFF. »Captured on Microtek 9800XL-SN:W4A36B00853 »Using Silver Fast 6.0 (Microtek) SM 9800XL software patched to V 6.4.0r9 on 2004-12-15 »set at Q-Factor 1.5 Screen 133 »DPI 200 »Scanner-> Internal:Calibration »Internal-> Monitor:Automatic »Internal-> Output: »Internal-> Adobe RGB (98) »Embedded Adobe RGB (98) icm (Photoshop 8.0 set the same) »Intent:Perceptual –Copyright Notice = USDA/ARS/NCRPIS - Iowa State University - Maize Staff –Date Created = 12/2/2002

NCRPIS Imaging Methods Hardware/Software Requirements –Intel Pentium III or 4 processor –10 GB of free disk space –512 MB of RAM –Microsoft Windows 2000 or XP –Adobe PhotoShop 7.0 or higher –Microsoft.NET Framework 1.1 (free download from Microsoft website) –Oracle client software –Internet connectivity to GRIN system in Beltsville

NCRPIS Imaging Methods GRIN Database Requirements 1)Add a new descriptor for each research crop to the descriptor table. The descriptor name (DQNAME) must be IMAGE (all upper case). 2)For each new descriptor added in step 1, add descriptor codes to the descriptor code table. Example codes for IMAGE of Amaranth: CodeDefinition. FIELDImages taken of plants in the field GHImages taken of plants in the greenhouse IN-SITUImages taken of plants in their adapted location PLANTImages of scanned plant samples SEEDImages of scanned seed samples

NCRPIS Imaging Methods GRIN Database Requirements (cont.) 3)For each descriptor code entered in step 2, add a matching evaluation entry to the evaluation table. IMPORTANT: The syntax of the evaluation name (ENAME) must have the exact same text as the descriptor code from step 2. It must also be the third word of the name with each word separated by periods. Example evaluation entries for Amaranth: Evaluation Name. AMARANTH.PHOTOGRAPH.FIELD AMARANTH.PHOTOGRAPH.GH AMARANTH.PHOTOGRAPH.IN-SITU AMARANTH.SCANNED.PLANT AMARANTH.SCANNED.SEED 4)To enter more than one image per Accession/Inventory/Observation combination you must add qualifier entries to the qualifier table. IMPORTANT: The syntax of the qualifier name (QUAL) must look like the following: Example evaluation entries for Zea: Qualifier Name. IMAGE-01 IMAGE-02 : IMAGE-99
