3. Brainstorm with your “like-minded” tablemates… As soon as the music starts, you have until the end of the song to complete three tasks: 1.Write down the types of assessments would you like to see in this class to meet your learning style. 2.Write down how you will be successful when studying and working in this class. 3.Prepare this as a brief presentation (write-up, skit, song….for the rest of your class (perhaps you can present it through your type of intelligence!)

4. Cell Visualization Procedure: 1.Put a drop of methylene blue on a slide. Caution: methylene blue will stain clothes and skin. 2.Gently scrape the inside of your cheek with the flat side of a toothpick. Scrape lightly. 3.Stir the end of the toothpick in the stain and throw the toothpick away. 4.Place a coverslip onto the slide 5.Use the SCANNING objective to focus. You probably will not see the cells at this power. 6.Switch to low power. Cells should be visible, but they will be small and look like nearly clear purplish blobs. If you are looking at something dark, dark purple, it is probably not a cell 7.Once you think you have located a cell, switch to high power and refocus. (Do NOT use the coarse adjustment knob at this point)

5. DNA Isolation Procedure: 1.Swish your mouth with your Gatorade for one minute. Chew lightly on your cheeks. Be careful not to swallow the solution. 2.Spit the solution back into the cup. 3.Carefully pour the spit solution into the plastic test-tube provided. Let your test tube rest on the desk top for at least 10 minutes. 4.After resting, you should have some cells settled in the bottom of your test tube. Use your pipette to carefully draw off most of the liquid on top, trying to avoid the cells in the bottom. Leave about 2 mL of liquid at the bottom. 5.Using your pipette, add 2mL of Cell lysis buffer to your cells/spit solution. Try to put the same amount of lysis buffer in that you have of spit solution. 6.Gently tilt the tube back and forth to mix the solution. Let it sit for 5 minutes.

DNA Isolation Procedure (continued): 7.Using the cold alcohol that Mrs. Akl provides, and holding your 15mL test tube at a 45⁰ angle, drip the cold alcohol down the side of the test tube, so that it forms a layer on top of your solution. 8.Let the tube sit upright and undisturbed for 5 minutes. 9.After 5 minutes, slowly invert the tube 5 times to help the DNA, which has begun to precipitate (or rise into the alcohol), to aggregate (or clump together). 10.With your pipette, carefully transfer the precipitated DNA along with about 1 ml of the alcohol solution into the small test tube that you will have for your necklace.
