CHAPTER 1: Some Tools of the Trade Lab 1.1 2014
2
Purpose of Lab 1.1 Become familiar with the small volumes of solutions used in molecular biology Introduce proper use of the micropipette Practice using the micropipette
3
Micropipettes
4
Parts of the micropipette
5
Micropipette use 1.Twist dial to desired volume 2.Add disposable pipette tip 3.Press plunger to first stop 4.To retrieve liquid, slowly release plunger 5.To transfer liquid, press plunger past first stop to second stop 6.Keep plunger down as you remove pipette 7.Eject tip

Micropipetting “nevers” Never use micropipette without a tip Never lay micropipette down with sample in the tip Never let the plunger button snap back Never set the P-20 pipette below 2.0μL or above 20.0μL
9
Pipetting techniques Hold micropipette and microfuge tubes at eye level when loading or dispensing samples
10
Pipetting techniques (cont.) Place both elbows on the table and use your other hand to stabilize the bottom of the pipette
11
Different amounts The amount you push the plunger depends on the amount: At 20.0μL the plunger is much higher than at 2.0μL
