Introduction to the Microscope Care Parts Focusing
3
Always carry with 2 hands Only use lens paper for cleaning Do not force knobs Always store covered Keep objects clear of desk and cords
4
Eyepiece Body Tube Revolving Nosepiece Arm Objective Lens Stage Stage Clips Coarse Focus Fine Focus Base Diaphragm Light
5
A. Place the slide on the microscope having the specimen in the center B. Use stage clips C. Click nosepiece to the lowest magnification (shortest = 4X) 1.Look into the eyepiece 2.Use the coarse adjustment knob FIRST to locate object
6
D. Rotate the nosepiece to the low power obj. = 10X 1.Look into the eyepiece 2.Use the course or fine adjustment knob to adjust the clarity The letter “e” Cotton fibers
7
E. Rotate the nosepiece to the high power obj.= 40X 1. Use the fine adjustment knob ONLY to bring the object into clearer vision
8
Calculating Total Magnification Each lens MULTIPLIES the magnification –The ocular lens (or eyepiece) magnifies 10X The Scanning (shortest) objective lens magnifies 4X The Low power objective lens magnifies 10X The High power objective lens magnifies 40X
9
QUIZ – 10 Questions 1. Email me your answers, answers only, at shestory@spsk12.net 2. Make sure you put you name on your email. What is the total magnification using the scanning objective lens? What is the total magnification using the high power objective? What is the total magnification using the low power objective? Once a slide is prepared and put into place on the stage, what is the correct order of objectives to use in order to bring the object on the slide into view with the most magnification?
10
QUIZ – con’t Which adjustment knobs are used with which objectives? If an objective was marked 100X, what is the total magnification possible with this lens? What part of the microscope holds the slide in place? What part holds and rotates the different Objectives? What parts do you hold onto to properly carry the microscope? What is your experience with using a microscope prior to this year? Explain what you remember looking at, if anything.
