3 Our microscopes have 3-4 objectives Each objective has the magnification power engraved on the side: 3.5x or 4.5: scanning objective 10x: low power objective 40x: high power objective 100x: oil immersion objective Take a slide from your box and test the magnification for each objective. You should have oil immersion in your table

4 Exercise A: Compound Light Microscope To focus use first the: Coarse-adjustment knob using the low power objective only! Then switch objective and focus using the fine-adjustment knob These microscopes are parafocal which means that the image stays in focus from one magnification power to the other. Take a slide from your box and test the magnification for each objective. You should have oil immersion in your table

5 Exercise A: Compound Light Microscope WHAT IS THE MAGNIFICATION OF YOUR OBJECTIVES? If your ocular is 10x and the objective is 3.5X the magnification is: 10 x 3.5=35 Calculate the magnification for: Scanning lens Low-power lens High-power lens Oil-immersion lens

6 Exercise A: Compound Light Microscope Find the diaphragm: regulates the light Using the same slide open and close the diaphragm to regulate the light.

7 Exercise A: Compound Light Microscope Grab it by the arm. Cleaning the lenses using the cleaning lens paper. Never use alcohol! When done, put the microscope in ‘resting position’. ALWAYS take care of your assigned microscope by: YOU ARE RESPONSIBLE FOR YOUR MICROSCOPE ANY THING THAT HAPPENS YOU NEED TO REPORTED. IF BREAKS IN YOUR WATCH YOU PAID FOR IT $800!

8 Exercise B: Dissecting Microscope This scope is to see larger things e.g. scale of a fish, the skin of a shark, the eye of an insect. There are several dissecting microscopes in your table
