Case study A 6-year-old male was brought to the emergency department after suffering burns to the right arm when a hot cup of coffee spilled on him. presented with bilateral partial thickness burns to the arm representing approximately 10% total body surface area (TBSA). The burn on the right arm was red, partial thickness wounds were pusy, with change of the skin color, and cellulitis, determine the causative micro-organisms, and antimicrobial therapy

Technique of collection Cleanse wound by removing excess debris from wound base with normal saline Thoroughly flush wound with sterile saline. Without touching swab to surrounding wound edges or skin, rotate tip of swab over a 1 cm area at the center of the open wound for 5 seconds Place swab in culture transport sleeve. Label culture collection/transport kit with name, birth date, specimen source, date and time of culture. Place in lab biohazard transport bag.

Technique of culture Remove protective cap from culture medium and insert cotton tipped applicator into the culture medium without contaminating the applicator shaft The streaking is done using a sterile an inoculation loop. The sample is spread across one quadrant of a petri dish containing a growth medium, usually an agar plate which has been sterilized in an autoclave Sterilize the loop in the flame.

Turn the plate 90 degrees and drag the steril loop through the area you have just streaked two to three times and continue to drag the loop in a "zig- zag" formation in the remaining half of the plate without touching that area again (area A). Sterilize the loop in the flame. Turn the plate 90 degrees. Repeat the procedure to make area B, C& zigzag being very careful not to touch any of the areas you previously streaked
