1 Isella et al., Supplemenrary Figure 1. ROC curve analysis for MACC1, MET and HGF gene dosage, as indicated

2 Isella et al., Supplementary Figure 2: Montecarlo simulation for the evaluation of significance of the chosen 5 th percentile MACC1 threshold. The histogram shows the distribution of 10’000 threshold values calculated as the 5 th percentile of random sets of 36 samples (black bars) versus the real 5 th percentile threshold calculated from the 36 poor prognosis samples MACC1 threshold value Real MACC1 threshold (p < 0.005) Random MACC1 thresholds Frequency

3 Supplementary figure in response to Reviewer #1 observation: “the AUC of 65 % that they demonstrate for MACC-1 is not that impressive. It would be interesting to see what this was for patients not undergoing neoadjuvant therapy.” The ROC curve illustrated in this figure has been obtained from patients not undergoing neoadjuvant therapy. Despite a lower AUC, the shape of the ROC curve remains interesting, i.e. it reaches high values on the y-axis for x-values around 0.6, as highlighted by the blue arrow. This means that low MACC1 levels are strongly associated with low recurrence probability.
