2 Image Enhancements Enhancements can be made for several reasons: Aesthetics Functionality Practicality If the image is modified, the process must be: Systematic Justifiable

3 Always make a copy of the image before processing!

4 Display Enhancement Use Look-Up Tables to adjust how pixels are displayed on the screen Non-destructive unless applied Doesn’t generally improve ability to analyse images

5 Filters Process individual pixels based on their environment Used to: enhance or reduce noise Pull out Edges Enhance morphological feature

6 Masks Create binary images for processing Can be used to extract further information from objects Use from Count/Size to fill holes or add smoothing

7 Colour Extraction Colour increases the complexity of analysis Colour isn’t always necessary or useful Colour images can be converted to mono from the Adjust ribbon by either: Extracting colour channels Averaging information from all channels

8 Colour Extraction Mono Average Colour Separation

9 Background Correction Uneven background can be aesthetically jarring and render analysis difficult Use background Correction to flatten backgrounds by: Subtracting background image Estimating background

10 Colour Composites Fluorescence imaging is typically monochrome Multiple images are taken separately from the same sample Colour composites recombine multiple monochrome images into a single workspace
